MONITORING OF INORGANIC POLYPHOSPHATE IN HUMAN ORGANISM AND ENVIRONMENT: THE REQUIREMENT AND APPROACHES |
N.A. Andreeva, T.V. Kulakovskaya, L.A. Ledova, L.P. Ryazanova, L.V. Trilisenko
The Ideas of I.S. Kulaev, pioneer in biochemistry and cell biology of inorganic polyphosphates, has now become widely accepted and developed in IBPM RAS. The evidences were obtained in favor of the important role of these polymers in the blood coagulation, inflammatory response, in bone tissue apatite formation, in transmission of nerve signals, in functioning of ion channels, and formation of amyloids. These properties of polyphosphates, as well as their widespread use in the food industry and other sectors of the economy, make relevant the development of highprecision methods for their analysis. The enzymatic method is based on the treatment of samples with specific exopolyphosphatases releasing phosphate, which is determined by standard colorimetry. Yeast exopolyphosphatases obtained from overproducing strains are an effective analytical reagent for highly specific and sensitive analysis of polyphosphates in biological samples. With the help of endopolyphosphatases, it is possible to obtain polyphosphates of a given length for research purposes and drug design.
