STRUCTURE-FUNCTION RELATIONSHIP IN THE YEAST PMA1 ATPASE

P-ATPases belong to a large class of ATP-phosphohydrolases widely spread from bacteria to humans. They use energy from ATP hydrolysis to transport a variety of cations and substrates and to maintain cell ion homeostasis. One of them are the fungal and yeast plasma membrane H+-ATPases, key enzymes in providing energy to support secondary solute transport systems and to maintain vitality of the cell. Studying these enzymes is of a top interest since fungi and yeasts are important and valuable object for basic and applied research and biotechnology. Studies, which have been carrying in IBPM RAS for years, have shed the new light on structure-function relationships in the yeast plasma membrane PМА1 H+-ATPase. By means of site-directed mutagenesis carried out in transmembrane segments M6 and M8, role of individual amino acid residues was revealed and peculiarities of the enzyme structure-function relationships were discovered, allowing formulating basic principles of mechanism of cation-transporting ATPases functioning. It has been shown that M6 segment is responsible for the cation transport specificity, while M8 segment regulates stoichiometry of transport. Using the presented data and crystal structure of the Са2+-АТРase, homology model was build allowing to postulate two proton (hydroxonium ion, Н3О+) binding sites in the PMA1 H+-ATPase, similarly to two Са2+ binding sites in the Са2+-АТРase of endoplasmic reticulum, thus pointing to the same mechanism of cation transport by these pumps.
